A first generation genetic linkage map of the European flat oyster Ostrea edulis (L.) based on AFLP and microsatellite markers ArchiMer
Lallias, Delphine; Beaumont, A.r.; Haley, C.s.; Boudry, Pierre; Heurtebise, Serge; Lapegue, Sylvie.
This study presents the first genetic linkage map for the European flat oyster Ostrea edulis. AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. Chi-square goodness of fit tests revealed high segregation distortion, which was significant for 32.8% of markers. Sixteen microsatellites and 235 AFLPs (170 AFLPs type 1:1 and 65 AFLPs type 3:1) were used to build sex-specific linkage maps using CriMap software. The first parental map (P1) consisted of 104 markers grouped in nine linkage groups, spanning 471.2 cM with an average spacing of 4.86 cM. The second parental map (P2) consisted of 117 markers grouped in ten linkage groups (which equals the haploid chromosome number),...

A first genetic linkage map of the blue mussel Mytilus edulis ArchiMer
Lallias, Delphine; Hecquet, Céline; Boudry, Pierre; Lapegue, Sylvie; Beaumont, Andy.
The blue mussel, Mytilus edulis, is widely distributed in the northern hemisphere, from the White Sea to the Atlantic coast of southern France, and is an economically important species (1 million tons per year, FAO, 2002). At present however, little domestication has been achieved with bivalves, in contrast to agricultural crops, livestock and horticultural species like tomatoes, where selection and production is entirely controlled by man. For bivalves, despite their economic importance, no lines with selected characteristics have been taken to the production scale. There is therefore a need to make up for lost time. Our interest in genetic mapping sprang from some results showing a genetic component to some economically important traits like growth and...

A multivariate approach to explore the genetic variability in the F2 segregating population of a tomato second cycle hybrid JBAG
Cabodevila,V.G; Picardi,L.A; Pratta,G.R.
Segregating progeny from the tomato Second Cycle Hybrids (SCH) that were obtained from crossing RIL (Recombinant Inbred Lines) allows the detection of new genetic combinations that could increase genetic variability in F2 populations. The objectives of the present study were to evaluate eleven tomato quality traits in a segregating F2 population obtained from a SCH and, then, to characterize the molecular diversity by six AFLP (Amplified Fragment Length Polymorphism) primer combinations. Different multivariate analyses were used to assess the degree of concordance among these two approaches to detect genetic variability. Sixty-nine F2 plants were obtained by selfing the SCH (ToUNR18xToUNR1). The parental RIL were derived from an interspecific cross between...

AFLP marker linked to water-stress-tolerant bulks in barley (Hordeum vulgare L.) Genet. Mol. Biol.
Altinkut,A.; Kazan,K.; Gozukirmizi,N..
The amplified fragment length polymorphism (AFLP) assay is an efficient method for the identification of molecular markers, useful in the improvement of numerous crop species. Bulked Segregant Analysis (BSA) was used to identify AFLP markers associated with water-stress tolerance in barley, as this would permit rapid selection of water-stress tolerant genotypes in breeding programs. AFLP markers linked to water-stress tolerance was identified in two DNA pools (tolerant and sensitive), which were established using selected F2 individuals resulting from a cross between water-stress-tolerant and sensitive barley parental genotypes, based on their paraquat (PQ) tolerance, leaf size, and relative water content (RWC). All these three traits were previously shown...

AFLP polymorphism in restored provenances of Ceiba aesculifolia within an urban heat island Phyton
Olvera-Mendoza,EI; Lara-Cabrera,SI; Sáenz-Romero,C; Lindig-Cisneros,R.
To quantify the effect on genetic diversity of restoring tree populations in an area under the influence of an urban heat island, we evaluated the genetic diversity of 72 Ceiba aesculifolia individuals, from a restoration experiment established from 2170 to 2260 m a.s.l. Reintroduced individuals were compared with the provenance from which the seeds were obtained, and two external provenances, for a total of 123 individuals. Samples were analyzed with three AFLP primer combinations. Polymorphisms of 38.4 to 62.5% were obtained. Genetic diversity estimated with the Simpson index ranged from 0.14 to 0.2. The provenances in the restoration site had higher diversity than the provenance that originated the seeds. Groups formed with provenances with similar...

AFLP-based genetic linkage maps of the blue mussel (Mytilus edulis) ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Hecquet, Celine; Boudry, Pierre; Beaumont, A.
We report the construction of the first genetic linkage map in the blue mussel, Mytilus edulis. AFLP markers were used in 86 full-sib progeny from a controlled pair mating, applying a double pseudo-test cross strategy. Thirty-six primer pairs generated 2354 peaks, of which 791 (33.6%) were polymorphic in the mapping family. Among those, 341 segregated through the female parent, 296 through the male parent (type 1:1) and 154 through both parents (type 3:1). Chi-square goodness-of-fit tests revealed that 71% and 73% of type 1:1 and 3:1 markers respectively segregated according to Mendelian inheritance. Sex-specific linkage maps were built with mapmaker 3.0 software. The female framework map consisted of 121 markers ordered into 14 linkage groups, spanning...

Agronomic and molecular analyses for the characterisation of accessions in Tunisian olive germplasm collections Electron. J. Biotechnol.
Taamalli,Wael; Geuna,Filippo; Banfi,Riccardo; Bassi,Daniele; Daoud,Douja; Zarrouk,Mokhtar.
In this study, a total of 14 agronomic traits, five AFLP primer combinations and ten SSR loci were used to describe and to classify a group of Tunisian olive genotypes into groups based on molecular profiles and agronomic traits. The analysis of variance of the agronomical data revealed significant differences among accessions for all measured traits. The mean phenotypic dissimilarity (0.34 with a range of 0.08-0.6) was low in comparison to dissimilarity calculated using AFLP (0.50 with a range of 0.16-0.70) and SSR markers (0.76 with a range 0.35-0.94). The correlation between the agronomical dissimilarity matrix and the matrices of genetic dissimilarity based on SSR and AFLP markers was very weak: 0.156 (p = 0.05) and 0.185 (p = 0.05), respectively. The...

Morello,Santiago; Giussani,Liliana M; Sede,Silvana M.

Analysis of genetic and epigenetic variation in in vitro propagated potato somatic hybrid by AFLP and MSAP marker Electron. J. Biotechnol.
Tiwari,Jagesh Kumar; Saurabh,Shaumaya; Chandel,Poonam; Singh,Bir Pal; Bhardwaj,Vinay.
Background: Genetic and epigenetic changes (DNA methylation) were examined in the tissue-culture propagated interspecific potato somatic hybrids between dihaploid Solanum tuberosum and S. pinnatisectum. Amplified fragment length polymorphism (AFLP) and methylation-sensitive amplified polymorphism (MSAP) were applied to detect the genetic and epigenetic changes, respectively in the somatic hybrids mother plants (1st cycle) and their regenerants (30th cycles sub-cultured). Results: To detect genetic changes, eight AFLP primer combinations yielded a total of 329 scorable bands of which 49 bands were polymorphic in both mother plants and regenerants. None of the scorable bands were observed in term of loss of original band of mother plant or gain of novel band...

Analysis of genetic population structure in Acacia caven (Leguminosae, Mimosoideae), comparing one exploratory and two Bayesian-model-based methods Genet. Mol. Biol.
Pometti,Carolina L.; Bessega,Cecilia F.; Saidman,Beatriz O.; Vilardi,Juan C..
Bayesian clustering as implemented in STRUCTURE or GENELAND software is widely used to form genetic groups of populations or individuals. On the other hand, in order to satisfy the need for less computer-intensive approaches, multivariate analyses are specifically devoted to extracting information from large datasets. In this paper, we report the use of a dataset of AFLP markers belonging to 15 sampling sites of Acacia caven for studying the genetic structure and comparing the consistency of three methods: STRUCTURE, GENELAND and DAPC. Of these methods, DAPC was the fastest one and showed accuracy in inferring the K number of populations (K = 12 using the find.clusters option and K = 15 with a priori information of populations). GENELAND in turn, provides...

Assessment of genetic diversity in sesame (Sesamum indicum L.) detected by Amplified Fragment Length Polymorphism markers Electron. J. Biotechnol.
Ali,Ghulam M; Yasumoto,Sirato; Seki-Katsuta,Masumi.
Sesame (Sesamum indicum L.) is one of the oldest oil crops and is widely cultivated in Asia and Africa. To determine the level of genetic diversity in relation to geographical origins and morphological characteristics, a total of 96 accessions have been collected from different parts of the world and were analyzed using AFLP techniques. Twenty-one primer pairs generated a total of 445 bands and among them 157 (35%) were polymorphic. Using UPGMA clustering analysis method based on the similarity coefficient, accessions were separated into two major groups. The first group mostly consists of Eastern Asian origin and another group consists of South Asian origin. Sub-clusters separated the accessions and form distinct diversity among groups. Considering the...

Assessment of silver-stained AFLP markers for studying DNA polymorphism in proso millet (Panicum miliaceum L.) Rev. Bras. Bot.
Karam,Décio; Westra,Philip; Niessen,Scott J.; Ward,Sarah M.; Figueiredo,José Edson Fontes.
Proso millet (Panicum miliaceum L.) is a serious weed in North America. A high number of wild proso millet biotypes are known but the genetic basis of its phenotypic variation is poorly understood. In the present study, a non-radioactive silver staining method for PCR-Amplified Fragment Length Polymorphism (AFLP) was evaluated for studying genetic polymorphism in American proso millet biotypes. Twelve biotypes and eight primer combinations with two/three and three/three selective nucleotides were used. Pair of primers with two/three selective nucleotides produced the highest number of amplified DNA fragments, while pair of primers with three/three selective nucleotides were more effective for revealing more polymorphic DNA fragments. The two better primer...

Association between molecular markers and behavioral phenotypes in the immatures of a butterfly Genet. Mol. Biol.
De Nardin,Janaína; Buffon,Vanessa; Revers,Luís Fernando; de Araújo,Aldo Mellender.
Abstract Newly hatched caterpillars of the butterfly Heliconius erato phyllis routinely cannibalize eggs. In a manifestation of kin recognition they cannibalize sibling eggs less frequently than unrelated eggs. Previous work has estimated the heritability of kin recognition in H. erato phyllis to lie between 14 and 48%. It has furthermore been shown that the inheritance of kin recognition is compatible with a quantitative model with a threshold. Here we present the results of a preliminary study, in which we tested for associations between behavioral kin recognition phenotypes and AFLP and SSR markers. We implemented two experimental approaches: (1) a cannibalism test using sibling eggs only, which allowed for only two behavioral outcomes (cannibal and...
